TP-FLIM雙光子熒光壽命成像光學造影劑
雙光子熒光壽命成像(Two-photon fluorescence lifetime imaging, TP-FLIM)技術集合了雙光子熒光成像和熒光壽命成像的優勢,能夠對生物組織進行高分辨三維成像的同時提供生物組織的生化特性信息,在**診療方面具有巨大的臨床應用前景。為了獲得較佳的雙光子熒光壽命成像效果,光學造影劑除了需要具有較強的雙光子吸收,用于降低激光的能量,避免高能量激光對組織的損傷;還需要具有穩定且較長的熒光壽命,用于去除生物自熒光對雙光子熒光壽命成像的干擾。然而,目前還鮮有材料能夠同時滿足上述要求。因此,迫切需要設計合成一種合適光學造影劑,用于推動雙光子熒光壽命成像技術在生物診療領域的發展。
一種基于熱激活延遲熒光(Thermally activated delayed ?uorescence, TADF)的有機光學造影劑,可用于雙光子活體熒光壽命成像。在該工作中,研究者充分利用熱激活延遲熒光材料通常具有較長延遲熒光(Delayed fluorescence)壽命這一優點,將具有聚集誘導熒光增強的有機熱激活延遲熒光材料包覆于兩親性共聚物內,成功制備了水溶性的有機納米顆粒。光學研究表明兩親性共聚物的包覆較大化了熱激活延遲熒光材料的聚集態,大大增加了材料的熒光壽命,獲得了可媲美磷光材料的超長發光壽命(4.2 μs)。更為重要的是,不像傳統磷光發光強度和壽命容易被氧氣猝滅,該材料對氧氣表現出明顯的惰性,即便在富氧區域仍然表現出穩定的熒光壽命,非常有利于獲得高信噪比的熒光壽命成像。同時,該熱激活延遲熒光材料由于具有Donor-Acceptor(D-A)結構,表現出良好的雙光子吸收截面,輕易實現了活體雙光子熒光壽命成像。
該研究為雙光子熒光壽命成像技術提供了一種良好的光學造影劑,對推動雙光子熒光壽命成像技術在臨床**診療中的發展具有重要意義。
